High-resolution model of Arabidopsis Photosystem II reveals the structural consequences of digitonin-extraction

Abstract In higher plants, the photosynthetic process is performed and regulated by Photosystem II (PSII). Arabidopsis thaliana was first plant with a fully sequenced genome, conferring it status of model organism; nonetheless, high-resolution structure its missing. We present Cryo-EM PSII supercomplex average resolution 2.79 Å, an important for future studies. The digitonin extracted complexes demonstrate importance of: LHG2630-lipid-headgroup in trimerization light-harvesting complex II; stabilization PsbJ subunit CP43-loop E DGD520-lipid; choice detergent integrity membrane protein complexes. Furthermore, our data shows at anticipated Mn 4 CaO 5 -site single metal ion density as reminiscent early stage photoactivation.
